大肠肿瘤中p53和bcl-2蛋白的表达
研究p53和bcl-2在大肠肿瘤发生中的作用。方法:应用免疫组化S-P法分别检测大肠正常粘膜、腺瘤及腺癌中p53和 bcl-2表达。结果:大肠腺癌p53表达率高于腺瘤组(P<0.05),p53表达与大肠癌的临床病理因素无关(P>0.05),bcl-2在正常粘膜基底部上皮细胞表达,在腺瘤(77.5%)和腺癌中(55%)bcl-2表达差异显著(P<0.01),高分化腺癌bcl-2表达率(73.7%)高于差分化腺癌(41.2%),在腺瘤和腺癌中bcl-2和p53呈负相关。结论:p53基因改变在大肠腺瘤癌变中起重要作用,大肠癌的发生与bcl-2基因在大肠腺瘤和腺癌中细胞凋亡抑制有关。

p53 and bcl-2 oncoprotein expression in colorectal tumor

ABSTRACT Purpose To study the role of p53、bcl-2 in colorectal tumorigenesis.Methods Using immunohistochemistry S-P method,the expression of p53 and bcl-2 were examined in normal colonic mucosas,hyperplastic polyps,adenomas and adenocarcinomas.Results The expression rate of colorectal adenocarcinomas was significant higher than that of adenomas (P<0.05).No significant correlation was observed between p53 expression and clinicopathological manifestations (P>0.05). bcl-2 was expressed in the basal epithelial cells of the lower colonic cypts in mucosa and hyperplastic polyps.Significant difference of bcl-2 expression was found between adenomas and adenocarcinomas(P<0.05).bcl-2 expression of well differentiated carcinomas was significant higher than that of poorly differentiated carcinomas (P<0.01).An inverse correlation was found between bcl-2 and p53 expression in adenomas and in adenocarcinomas (P<0.01).Conclusion Alteration of p53 gene is involved mainly in the transformation from benign adenoma to adenocarcinoma, colorectal carcinogenesis is related to the inhibition of apoptosis.
